Hve you displayed your structure? In the x-direction the structure is continuous wit four-membered rings between the cells. In the y-direction the structure is discontinuous with the gap of 4A between the cells. Is this what you want to calculate?
Realaxation of a large cell runs well with just single k-point.

Thank you for your answer... I would like to calculate or to do a geometric optimization using a graphene layer...

At least, in my point of view, the c-axes is too small for "single-layer" simulation (of cource, if you want to work on several layers structure, you'd better using graphite symmetry, P63mc space group). Normally, the "vacumm slab" for graphene layer calculations is 10~20 Angstrom.
Furthermore, since graphene-like structure is hexagonal, why not set a0 and a1 vectors as: a0*(1.0, 0.0, 0.0) and a1*(-0.5, sqrt(3)/2, 0.0) ?
I am not sure whether there are something special in VASP, but based on common ab-initio knowledge, using hex-coordinate based model is better in graphene layer field to achieve the better 3D periodicity expansion.
